when the option On Fly (jQuery) is on, the flags, drop down list appears but when is clicked, it doesnt work

Check for javascript errors. May be you already have another jQuery library loaded on your page. Try to disable it in GTranslate module. If that doesn't help, try to disable it in the other extension which loads jQuery. If there is no option for that, contact the other extension developers and ask them to make it compatible with GTranslate.

Hi, I installed the plugin on my wordpress and do not know how to use it, I see no change in the front, Please can you tell me how it works?

You need to add GTranslate widget into your sidebar, so you can see it on the wordpress front-end.
It can be configured from Settings -> GTranslate
